From Barnes & Tuffrey (1973). cell itself, but against an NZB red cell/NZB autoantibody complex (Fig. 30) (Barnes & Tuffrey, 1973). A breakdown of tolerance in the NZB^CFW chimaeras (Barnes & Tuifrey, 1972Ò) is supported by other changes that occurred as the animals aged. In one, the CFW red cell population disappeared, while another lost its NZB red cells and some of the black hairs in the coat, and acquired the ability to reject NZB skin (Barnes & Tuffrey, 1972a). These shifts in cell populations need not, however, have had an immunological basis. Serum haemolytic complement activity was absent (Barnes et al, 1974c) as in the NZB strain, although CFW mice have normal levels of complement and chimaeras of strain combinations not involving NZB showed little or no reduction in com¬ plement activity. Deposits of complement were later detected in renal lesions, together with antibody. The pattern of immunofluorescent staining in the lesions closely resembled that seen in acute graft-versus-host disease. Excess removal of complement in complexes is known to accompany graft-versus- host disease, and could explain the absence of serum complement activity. A graft-versus-host assay based on popliteal lymph node weight gain proved positive. Immunofluorescence techniques (Barnes et al., \91Aa) not only established the presence of both cell populations in thymus, spleen, lymph nodes and kidney of the chimaeras, but also revealed the phagocytosis of erythrocytes by monocytes and macrophages in all four animals.
